The collaboration marks a push to develop a differentiated approach to obesity treatment by targeting fat tissue directly
Rona Therapeutics has struck a major licensing deal with Roche, granting the Swiss pharmaceutical giant exclusive worldwide rights to an experimental RNA interference (RNAi) therapy targeting obesity and metabolic diseases in an agreement that could be worth more than $1.16 billion.
The deal centres on Rona's investigational candidate targeting activin receptor-like kinase 7 (ALK7), a protein predominantly expressed in fat tissue. The therapy uses Rona's proprietary extrahepatic delivery platform, designed to deliver RNAi treatments beyond the liver.
Under the agreement, Rona will receive upfront and near-term milestone payments of up to $100 million and is eligible for up to $1.06 billion in additional development, regulatory and commercial milestone payments, alongside tiered royalties on global net sales.
The collaboration marks a push to develop a differentiated approach to obesity treatment by targeting fat tissue directly, potentially complementing existing incretin-based therapies.
ALK7, encoded by the ACVR1C gene, is a genetically validated target for obesity and metabolic diseases. Inhibiting the protein directly in fat cells could selectively reduce visceral fat and other ectopic fat deposits while preserving lean mass — a potential advantage in a treatment landscape increasingly focused on the quality, as well as the amount, of weight lost.
"Obesity is a complex chronic disease with significant unmet medical needs for new mechanisms that can improve the quality and durability of weight loss," said Stella Shi, CEO and Founder of Rona Therapeutics.
"ALK7 is a compelling obesity and metabolic disease target with the potential to directly address adipose tissue biology. Rona Therapeutics is committed to discover and develop novel RNAi therapies to benefit patients with obesity and metabolic diseases globally. This collaboration combines Rona's pioneering extrahepatic RNAi R&D platform with Roche's strong expertise and commitment in global development and commercialization of medicines for the treatment of obesity and metabolic diseases to bring this important therapeutic candidate to patients."
For Roche, the agreement strengthens its push into next-generation treatments for obesity and cardiometabolic, renal and related conditions, adding a potential new mechanism to its existing pipeline.
"Obesity represents a significant health challenge that requires differentiated treatment options for patients," said Boris L. Zaïtra, Head of Corporate Business Development at Roche. "We are excited to partner with Rona Therapeutics, complementing our robust CVRM pipeline and reinforcing our commitment to delivering next-generation metabolic therapies."
The agreement gives Roche exclusive worldwide rights to the investigational candidate, while Rona stands to benefit from substantial milestone payments and royalties if the programme advances through development and achieves commercial success.
The therapy remains investigational, and its potential to deliver meaningful clinical benefits will depend on further development and evaluation. However, the deal underscores growing industry interest in approaches that target the underlying biology of obesity beyond conventional weight-loss mechanisms.
